| I. Background | |
| CRF1R 屬于 B 類 GPCR 家族,在中樞神經系統(下丘腦、杏仁核、前額葉皮層)及外 周組織(胃腸、皮膚)表達,是機體應激應答的核心調控者。CRF1R 的內源性配體促腎上 腺皮質激素釋放因子(CRF)和 尿皮質素 1(UCN1)結合后,觸發受體構象變化,優先偶 聯 Gs 蛋白,激活腺苷酸環化酶(AC),升高 cAMP,啟動 PKA/CREB 信號軸,驅動下丘 腦-垂體-腎上腺軸(HPA 軸)激活,最終促進 糖皮質激素(皮質醇)釋放以應對生理/心理 壓力;同時通過β-arrestin 通路調節受體脫敏內化。該受體過度激活與焦慮癥、抑郁癥、腸 易激綜合征(IBS)及藥物成癮密切相關。 | |
