| I. Background | |
| CRF1R 屬于 B 類 GPCR 家族，在中樞神經系統（下丘腦、杏仁核、前額葉皮層）及外 周組織（胃腸、皮膚）表達，是機體應激應答的核心調控者。CRF1R 的內源性配體促腎上 腺皮質激素釋放因子（CRF）和 尿皮質素 1（UCN1）結合后，觸發受體構象變化，優先偶 聯 Gs 蛋白，激活腺苷酸環化酶（AC），升高 cAMP，啟動 PKA/CREB 信號軸，驅動下丘 腦-垂體-腎上腺軸（HPA 軸）激活，最終促進 糖皮質激素（皮質醇）釋放以應對生理/心理 壓力；同時通過β-arrestin 通路調節受體脫敏內化。該受體過度激活與焦慮癥、抑郁癥、腸 易激綜合征（IBS）及藥物成癮密切相關。 | |
