| I. Background | |
| 5-羥色胺受體(5-HTR)家族是介導血清素(5-羥色胺)信號傳遞的關鍵受體群,分為 7 大類(5-HT1-7)共 14 個亞型,包含 G 蛋白偶聯受體(GPCR) 和配體門控離子通道兩 類結構,廣泛分布于中樞神經系統、外周組織及血小板,通過多維度調控情緒、認知、心 血管、胃腸功能及疼痛感知。 5-HTR2C 受體作為 Gq 蛋白偶聯的 5-羥色胺受體亞型,在中樞神經系統(尤其下丘腦、 邊緣系統及脈絡叢)高表達,以內源性配體 5-羥色胺(5-HT) 激活后觸發 PLC-IP3/DAG- Ca 2+信號軸,通過調節神經元興奮性與神經遞質釋放(如抑制多巴胺能通路)深度參與食 欲調控、情緒穩定性及癲癇閾值維持。5-HTR2C 功能受獨特的 RNA 編輯機制調控,臨床 研究發現該受體基因(HTR2C)的功能缺失性突變與暴食癥、抗抑郁藥耐藥性抑郁癥及嬰 兒癲癇綜合征(Dravet 綜合征) 強相關,而選擇性激動劑(如 lorcaserin) 曾用于肥胖治 療(通過激活下丘腦 POMC 神經元增強飽腹感),反向激動劑(如 SB-242084) 則作為潛 在抗抑郁靶點進入Ⅱ期試驗,凸顯其作為神經精神代謝疾病關鍵節點的雙重治療價值。 |
